The Three Rs framework places reduction of animal use at its center, and significant progress has been made in developing validated alternatives that eliminate the need for animal testing in some research areas.
Every animal use eliminated through validated alternatives represents prevented suffering without loss of scientific information. The development of organ-on-a-chip platforms that replicate human tissue responses, computational toxicity prediction models, and in vitro systems that outperform animal models in predicting human outcomes represents genuine progress. The primary barrier to adoption is regulatory — validated alternatives must be accepted by agencies before they can replace required animal tests. Accelerating regulatory acceptance of validated alternatives, particularly for established testing requirements, is the highest-leverage intervention for reducing laboratory animal numbers and the associated welfare costs.
