Laboratory rodent welfare involves both procedural harms (injections, surgeries, induced disease models) and baseline housing welfare. Single housing of social mice causes measurable stress responses including elevated corticosterone, immune suppression, and stereotypic behavior — and paradoxically compromises experimental validity by introducing a major confound. Nesting material provision, a simple and cheap refinement, reduces cold stress, improves welfare, and reduces inter-individual variation. Pain recognition in mice has historically been poor; the Mouse Grimace Scale, developed from orbital tightening and ear/whisker position, allows objective assessment. The welfare of genetically modified animals requires specific monitoring protocols given unpredictable phenotypic effects. Institution-wide refinement programs led by Institutional Animal Care and Use Committees (IACUCs) drive systematic improvements.
