The 3Rs framework has driven the most significant welfare improvements in laboratory animal care over the past six decades. Replacement — eliminating animal use — is the most impactful principle but the hardest to achieve for whole-system research questions. Progress in organoids, organ-on-chip systems, and computational biology is accelerating replacement in toxicology and some disease areas. Reduction requires careful study design and statistical planning to extract maximum data from minimum animals. Refinement — improving the lives of animals still used — encompasses everything from analgesic protocols and humane endpoints to environmental enrichment, social housing, and low-stress handling training. Institutional Animal Care and Use Committees (IACUCs) and ethics boards serve as gatekeepers for 3Rs implementation, though quality varies considerably between institutions.
