The 3Rs provide an ethical framework that has meaningfully reduced laboratory animal suffering while enabling continued scientific progress. Replacement technologies — from cell cultures to organ-on-a-chip systems — are increasingly valid for many research questions. Reduction requires rigorous statistical planning that many studies still lack. Refinement encompasses pain management, housing enrichment, and humane endpoints. Despite progress, implementation is uneven across research institutions and countries, and investment in alternative methods development remains insufficient relative to need.
