Progress and challenges in reforming laboratory animal welfare, including advances in the 3Rs framework, non-animal alternatives, regulatory improvements, and remaining gaps.
Laboratory animal welfare reform has made genuine progress over the past two decades, driven by improved regulatory frameworks, advances in non-animal alternatives technology, and growing scientific and public attention to the welfare of research animals. The EU Directive 2010/63/EU represents the most comprehensive mandatory welfare framework, requiring project authorization, species justification, severity classification, and post-procedure care standards that significantly exceed requirements in many other jurisdictions. The FDA Modernization Act 2.0 in the US is a landmark regulatory change that removes mandatory animal testing as a precondition for human trials — potentially accelerating both drug development and animal use reduction simultaneously. Non-animal alternatives including organoids (miniature organs grown from human cells), organ-on-chip microfluidic systems, and artificial intelligence toxicology prediction models are advancing rapidly, with some already providing better predictive accuracy than animal models for specific applications. Despite this progress, significant welfare improvements remain unrealized: pain management in laboratory animals remains inconsistent; housing conditions for social species frequently violate known welfare needs; and the enormous scale of use in some jurisdictions occurs without adequate oversight or transparency.
