The Three Rs framework has substantially improved laboratory animal welfare since its development, embedding welfare considerations into research design and institutional review. However, implementation gaps persist: replacement technologies are advancing but cannot yet substitute for all whole-organism research; reduction goals are sometimes undermined by statistical under-powering that wastes animal lives; and refinement improvements vary greatly by institution and species. The increase in transgenic mouse use has created new welfare challenges not fully addressed by existing frameworks.
